    Experimental demonstration of RGB LED-based optical camera communications

    Red, green, and blue (RGB) light-emitting diodes (LEDs) are widely used in everyday illumination, particularly where color-changing lighting is required. On the other hand, digital cameras with color filter arrays over image sensors have been also extensively integrated in smart devices. Therefore, optical camera communications (OCC) using RGB LEDs and color cameras is a promising candidate for cost-effective parallel visible light communications (VLC). In this paper, a single RGB LED-based OCC system utilizing a combination of undersampled phase-shift on off keying (UPSOOK), wavelength-division multiplexing (WDM), and multiple-input multiple-output (MIMO) techniques is designed, which offers higher space efficiency (3 bits/Hz/LED), long-distance, and nonflickering VLC data transmission. A proof-of-concept test bed is developed to assess the bit-error-rate performance of the proposed OCC system. The experimental results show that the proposed system using a single commercially available RGB LED and a standard 50-frame/s camera is able to achieve a data rate of 150 bits/s over a range of up to 60 m

    Functional characterization of cellulases identified from the cow rumen fungus Neocallimastix patriciarum W5 by transcriptomic and secretomic analyses

    <p>Abstract</p> <p>Background</p> <p><it>Neocallimastix patriciarum</it> is one of the common anaerobic fungi in the digestive tracts of ruminants that can actively digest cellulosic materials, and its cellulases have great potential for hydrolyzing cellulosic feedstocks. Due to the difficulty in culture and lack of a genome database, it is not easy to gain a global understanding of the glycosyl hydrolases (<it>GHs</it>) produced by this anaerobic fungus.</p> <p>Results</p> <p>We have developed an efficient platform that uses a combination of transcriptomic and proteomic approaches to <it>N. patriciarum </it>to accelerate gene identification, enzyme classification and application in rice straw degradation. By conducting complementary studies of transcriptome (Roche 454 GS and Illumina GA IIx) and secretome (ESI-Trap LC-MS/MS), we identified 219 putative <it>GH </it>contigs and classified them into 25 <it>GH</it> families. The secretome analysis identified four major enzymes involved in rice straw degradation: β-glucosidase, endo-1,4-β-xylanase, xylanase B and Cel48A exoglucanase. From the sequences of assembled contigs, we cloned 19 putative cellulase genes, including the <it>GH1</it>, <it>GH3</it>, <it>GH5</it>, <it>GH6</it>, <it>GH9</it>, <it>GH18</it>, <it>GH43 </it>and <it>GH48 </it>gene families, which were highly expressed in <it>N. patriciarum </it>cultures grown on different feedstocks.</p> <p>Conclusions</p> <p>These <it>GH </it>genes were expressed in Pichia pastoris and/or Saccharomyces cerevisiae for functional characterization. At least five novel cellulases displayed cellulytic activity for glucose production. One β-glucosidase (W5-16143) and one exocellulase (W5-CAT26) showed strong activities and could potentially be developed into commercial enzymes.</p

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals &lt;1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    Genome-wide association study of lung adenocarcinoma in East Asia and comparison with a European population.

    Lung adenocarcinoma is the most common type of lung cancer. Known risk variants explain only a small fraction of lung adenocarcinoma heritability. Here, we conducted a two-stage genome-wide association study of lung adenocarcinoma of East Asian ancestry (21,658 cases and 150,676 controls; 54.5% never-smokers) and identified 12 novel susceptibility variants, bringing the total number to 28 at 25 independent loci. Transcriptome-wide association analyses together with colocalization studies using a Taiwanese lung expression quantitative trait loci dataset (n = 115) identified novel candidate genes, including FADS1 at 11q12 and ELF5 at 11p13. In a multi-ancestry meta-analysis of East Asian and European studies, four loci were identified at 2p11, 4q32, 16q23, and 18q12. At the same time, most of our findings in East Asian populations showed no evidence of association in European populations. In our studies drawn from East Asian populations, a polygenic risk score based on the 25 loci had a stronger association in never-smokers vs. individuals with a history of smoking (Pinteraction = 0.0058). These findings provide new insights into the etiology of lung adenocarcinoma in individuals from East Asian populations, which could be important in developing translational applications

    [[abstract]]Skin whitening, tumor, slimming and acne disease are the significant subjects of dermatology. Melanocortin 1 receptor (MC1R) and tyrosinase are the important targets for whitening. RAS family, B-RAF and cyclin-dependent kinase 2 (CDK2) are the essential targets for skin tumor. Adenosine monophosphate-activated protein kinase (AMPK) and Glucagon-like Peptide-1 receptor (GLP-1) are the main targets for obesity disease. Salt inducible kinase 1 (SIK1) is the critical target for acne disease. In the recent years, molecular modeling is undoubtedly an emerging technique to investigate many kinds of diseases due to rapid development of computer hardware and software. This research utilized the concept of computer-aided drug design with molecular dynamics (MD) simulation to explore the secret of dermatologic diseases. Pure traditional Chinese medicine (TCM) database is useful for accurate application of herbal medicine to explore major dermatologic disorders. This study used virtual screening to predict that merresectine C, abrine / aknadicine or 16beta-hydroxy-19s-vindolinine N-oxide / tetrahydropalmatine, reserpiline or (+)-corydaline, eugenyl_beta-D-glucopyranoside or 6-O-cinnamoyl-D-glucopyranose / wenyujinoside or 28-deglucosylchikusetsusaponin IV, platyphyllonol or meso-hannokinol could bind to target protein successfully and stably. Quantitative structure-activity relationship (QSAR) predictive models such as support vector machine and multiple linear regression were used to verify these screened pure TCM compounds to be the potent lead medicine.     [[abstract]]Adenosine monophosphate-activated protein kinase (AMPK) acts as a master mediator of metabolic homeostasis. It is considered as a significant millstone to treat metabolic syndromes including obesity, diabetes, and fatty liver. It can sense cellular energy or nutrient status by switching on the catabolic pathways. Investigation of AMPK has new findings recently. AMPK can inhibit cell growth by the way of autophagy. Thus AMPK has become a hot target for small molecular drug design of tumor inhibition. Activation of AMPK must undergo certain extent change of the structure. Through the methods of structure-based virtual screening and molecular dynamics simulation, we attempted to find out appropriate small compounds from the world's largest TCM Database@Taiwan that had the ability to activate the function of AMPK. Finally, we found that two TCM compounds, eugenyl_beta-D-glucopyranoside and 6-O-cinnamoyl-D-glucopyranose, had the qualification to be AMPK agonist
